제목 Comparison of Gene Expression between Bone Marrow Derived mesenchymal Stem Cells and Adipose Derived Stem Cells Using Polyethylenimine
초록 Bone marrow mesenchymal stem cells (BMSCs) and adipose stem cells (ADSCs) can migrate into injured site with target gene. Here, we tried to use Polyethylenimine (PEI) for nonviral vector system to apply each stem cell and compared gene transfection efficiency. Physical property of nanoparticles was about 133-205 nm particle size and 28-43 mV of surface charge at N/P 4-16. Cytotoxicity was increased by the amount of PEI. FACS analysis and fluorescent microscope data was shown the expression of green fluorescent protein at N/P 8 and 16 were more effective than lipofectamine as reference carrier. Furthermore, ADSCs were so easy to uptake nanoparticles that ADSCs expressed more effective than BMSCs. However, N/P 16 has cytotoxicity compared with lipofectamine so N/P 8 can be optimum to use on BMSCs and ADSCs. Conclusion PEI used in gene carrier into BMSCs and ADSCs is effective on nonviral vector system and ADSCs are more effective compared with BMSCs.
