Chemical Stabilization of Self-assembled Core-shell Micelles of Multiblock Polypeptides for Efficient Drug Delivery |

Chemical stabilization of core-shell polymer micelles has been widely studied for efficient drug delivery. We report that multiblock polypeptides composed of thermally responsive elastin-based polypeptide (EBP) diblock, Cys-incorporated block and biologically active domain were self-assembled into the core-shell micelles above lower critical solution temperature and their shell was chemically crosslinked via UV irradiation or oxidation. The acrylic moieties were introduced into the hydrophilic EBP block of the shell and then chemically stabilized by UV exposure once thermally-triggered micelles were formed. On the other hand, reversible oxidation and reduction of the Cys block of the shell was achieved by using oxidizing and reducing agents. Shell crosslinking of the self-assembled core-shell micelles was characterized by dynamic light scattering and electron microscopy. These stabilized polypeptide nanocarriers would be useful for controlled drug delivery systems. |
