제목 Heterologous expression of CYP102D1 in Escherichia coli and enhancement of catalytic efficiency by the construction of artificial self-sufficient cytochrome P450
초록 Among 33 cytochrome P450s in Streptomyces avermitilis MA4680 which are cloned, SAV 7469(CYP105D7) shows the highest catalytic activity with daidzein under the reaction with camA as ferredoxin and camB as ferredoxin reductase. Also, SAV 575(CYP102D1) is found to be the only self-sufficient cytochrome P450 in S.avermitilits. The artificial self-sufficient cytochrome P450 is constructed by employing CYP102D1. This P450 system is based on the substitution of P450 domain in CYP102D1 with CYP105D7 and expected to achieve increased enzymatic activity. Biotransformation of daidzein catalyzed by this enzyme using E.coli and S.avermtilis as host systems is confirmed with GC/MS and HPLC analysis.
