Activated macrophages are a major ingredient of the inflammatory system. In inflammation, macrophage have a critical role; initiation, maintenance, and resolution. Activated macrophages, overexpressing scavenger receptor type A (SR-A), are the attractive therapeutic target for atherosclerosis and rheumatoid arthritis (RA). Dextran sulfate (DS) acts as a ligand for SR-A. In this study, we made SR-A targeting materials. First, the hydrophilic diblock copolymer, composed of DS and poly(glycerol monomethacylate), was prepared as the coating material to modify surface of the superparamagnetic iron oxide nanoparticles (SPION). DS-coated SPION, which specifically binds to the receptor on activated macrophages of the atherosclerotic plaque. Second, in an attempt to develop self-assembled nanoparticles for targeting RA, we prepared the amphiphilic copolymer via click chemistry, composed of the hydrophilic DS as the ligand for SR-A and polycaprolactone as the hydrophobic segment. |
