Zwitterionic fluorescent nanoparticles are developed by using polysulfobetaine methacrylate, NIPAAm and BODIPY fluorophores to identify the cancer cells in response to intra and extracellular stimuli. The nanoparticles were well characterized by H-NMR, UV-vis, fluorescence spectra, and DLS measurement. The fluorescent nanoparticles exhibit minimal fluorescence intensity at physiological temperature and pH, while strong emissions are observed at acidic and basic environment. Confocal images of the nanoparticles in MDA-MB 231 cells also confirm the intense green fluorescence at low and high pH values but no fluorescence signal under physiological pH. The intracellular co-localization of the nanoparticles, using LysoTracker Blue, showed the green emission of nanoparticles and the blue emission of LysoTracker Blue co-localize and overlay very well with each other which confirmed the localization of fluorescent nanoparticles. |
