Gold nanorods (AuNRs) can used for photothermal therapy (PTT) under infrared (NIR) light exposure. In this study, we synthesized the AuNRs immobilized with small interfering RNA (siRNA). AuNRs are centrifuged to remove excess amount of cetyltrimethylammonium bromide (CTAB) because CTAB is well-known toxic cationic surfactant. To stabilize the AuNRs, thiol–oligonucleotide (HS–T10–overhang) was added on the surface of AuNRs as well as thiol–polyethylene glycol–amine (HS–PEG–NH2). siRNA can be later hybridized by overhang base-paring with immobilized oligonucleotide. With AuNR–siRNA conjugates, we can expect to achieve effective endosomal releasing under laser stimulation and photo–triggered gene silencing can be verified in the green fluorescence protein (GFP) expressing KB cells. |
