Co-delivery of BMP2 and SOX9 to recover chondrogenic property of dedifferentiated chondrocyte |

Bone morphogenic protein (BMP) is an important signal to upregulate osteogenesis and chondrogenesis of stem cell. SRY-related HMG-box gene 9 (SOX9) was reported as the key transcription factors of chondrogenesis. We hypothesized that the co-delivery of the BMP2 and SOX9 genes would render dedifferentiated chondrocyte to recover normal condrogenic properties efficiently. To aim this, we constructed a bicistronic vector encoding the BMP2 and SOX9 genes linked to the 2A peptide sequence. Therefore we confirmed above 70% transfection efficiency of BMP2 and SOX9 gene by RT-PCR and western blot analysis. In RT-PCR analysis and alcian blue staining, expression of chondrogenesis-related markers and GAG matrix formation were significantly increased when BMP2/SOX9 was delivered simultaneously compared to BMP2 or SOX9 gene. These findings demonstrate that the chondrogenic lineage-determining two genes would be able to enhance chondrogenic re-differentiation of dedifferentiated chondrocyte. |
