In this study, we designed MNPs not only for high magnetic resonance (MRI) contrast enhancement but also for excellent colloidal-stability and targeting capabilities. Preparing the monodisperse MNPs with uniform size (7-10 nm) and defined morphology. Furthermore, we fabricated amphiphilic lipopolymer consisted with phosphatidylethanolamine-b-hyperbranched polyglycidol (PE-b-HBPG). After that, we conjugated folic acid (FA) on to the PE-b-HBPG for targeting tumor cells which have the ability to over express the folate receptor in cancer cells. Finally, MNPs were labeled with this PE-b-HBPG-FA by ligand exchange reaction to make MNPs soluble in water and increase targetability through receptor-mediated cellular binding and uptake. We found HCT116 colon cancer cells have over expressed folate receptor in the other hand, RKO colon cancer cells didn’t have folate receptor. As a result, only confocal image of settled HCT116 shows the fluorescence because of MNPs labelled with folic acid. |
