Simvastatin/BCP/PCL Composite Coating on BCP-ZrO2 Scaffold for Drug Delivery System and Its Characterization |

Porous bone substitute, which have interconnected macro pores, has been focused on improving their mechanical and functional properties. Especially, it is highly required to develop drug-entrapped porous bone substitute for faster early bone formation as well as controlling their material properties such as bioresorption rate. In this work, porous BCP-ZrO2 scaffold was coated with simvastatin-BCP-PCL composite. With alteration in the coating concentration and BCP/PCL ratio, the morphology, mechanical properties, and in vitro biodegradable were investigated. Increasing BCP/PCL ratio made the coating surface to be rough. However, for all concentrations and compositions, uniform coatings were formed with the BCP particles being dispersed homogeneously in the PCL phase. With the composite coating, the mechanical properties were improved by several orders of magnitude. The in vitro degradation of the composite coatings increased linearly with incubation time and the rate differed with the coating concentration and the BCP/PCL ratio. At a short period (< 6h), 10-20% simvastatin drug that coating on surface was released. However, in vitro study showed that the release rate was sustained for prolong periods and highly dependent on the degree of coating dissolution. |
