Halogenated arenes are widely used for pharmaceuticals and fine chemicals. Flavin-dependent halogenases are known for halogenation of arenes with specific regioselectivity. Among various halogenated compounds, dichloroindigo is known for highly valuable products due to its color and electrical conductivity, synthesized by dimerization of two oxidized chloroindole. We are studying halogenase from Streptomyces species to make chloroindole from indole. However, halogenase itself showed low activity because of its low solubility within cell. We fused the halogenase with flavin reductase(Fre), FAD regeneration enzyme, in order to solve the solubility issue and obtain the advantages of fusion. As a result, fusion-halogenase showed enhanced solubility. We further expect the activity of halogenase and Fre fusion enzyme would be enhanced. |
