The Effect of Poly(L-lactide-co-glycolide) Microsphere in Gellan Gum Sponge on Proliferation of Annulus Fibrous Cells |

Abstract: Gellan gum(GG) is good heat resistance and acid resistance enzymes thus used widely. In this work, to improve mechanical properties of GG sponge, GG sponges were mixed with poly(lactic-co-glycolic acid (PLGA)) microsphere(PM) and then seeded cells. We fabricated a GG sponge with various contents (10, 30, 50 and 80wt% of GG weight) of PM. The various contents of PM/GG sponge were characterized by SEM, FT-IR, water uptake and degradation. MTT assay, GAG assay and immunohistochemistry staining were carried out to confirm cell viability and effect of ECM secretion from cells in the PM/GG sponges. The results confirmed that 50 wt% PM/GG sponge showed steady increasing proliferation rate. When microspheres were loaded too much in the PM/GG sponge, cell proliferation was disturbed because there is not enough pore-size and porosity for proliferation of cells. The AF proliferation in 50 wt% PM/GG sponge showed very good. This research was supported by WCU(R31-20029) and MBC(A040003). |
