The Complex of Polycation Natural Polymer with pDNA for Gene Delivery System: in vitro study |

Chitosan and PEI well known natural polymer have been widely experimented for gene delivery system as non-viral vector. In the present study, chitosan and PEI were conjugated by coupling agent such as EDC and NHS. Characterization of chitosan/PEI complex was measured by 1H-NMR and FT-IR. Target ligand was modified to chitosan/PEI complex for target gene delivery to specific cancer cell. Particle size and morphology of target ligand modified chitosan/PEI complex were monitored by DLS and TEM. Gel retardation assay was also performed to confirm the pDNA protecting efficiency, pDNA release efficiency and binding efficiency. When, the increased PEI contents of target ligand modified chitosan/PEI complex, binding efficiency was increased at low complex ratio. |
