Several transplantation of retinal pigment epithelium (RPE) cells can causes degeneration. The transplantation of cell in human needs a suitable scaffold. In the present study, we prepared the gellan gum films and investigated to determine their suitable films for ARPE-19 cells culture. Gellan gum is widely used in biotechnology because it forms a transparent gel which is heat resistant and its strength is less dependent on pH in comparison with other polysaccharide gels. We fabricated biomaterial gellan gum film (1, 2, 3 and 4 % content of gellan gum) and ARPE-19 cells were seeded on gellan gum films. In the result of MTT assay, cell viability on 3% gellan gum films was higher than other content films. specific mRNA expression of ARPE-19 cells and protein are observed on gellan gum films. Gellan gum films provide suitable surface for ARPE-19 cells and suitable gellan gum content affect to improve the retinal transplant.
