제목 | Design of functional nano-bio interfaces using CutA1 as a proteinaceous scaffold |
초록 | Technologies to detect and control interactions between biomolecules are used in various fields. In particular, the importance of functional molecular recognition surfaces (nano-bio interfaces) where proteins, such as antibodies and artificial antibody-like proteins are immobilized to interact with target molecules, has been widely recognized. CutA1 is a homotrimeric protein with the highest structural stability reported so far, and its characteristics allows CutA1 to tolerate insertion of ligand biomolecular sequences, which is not the case for other proteins. We have shown that the orientation of ligand biomolecules displayed on CutA1 can be controled to exhibit better functionality after immobilization. In this research, we designed new molecular recognition elements by utilizing single-stranded CutA1 or inserted PPI pair (Barnase-Barstar) to CutA1. Then, the charracteristics of nano-bio interfaces after their immobilization were examined. |
