제목 Antioxidant and anti-apoptotic effect of vanillin-incorporated copolyoxalate nanoparticles in ischemia/reperfusion injury
초록 The major culprit in pathogenesis in ischemia/reperfusion (I/R) injury involves damage from ischemic stroke, inflammation cascade and overproduction of reactive oxygen species (ROS). Particularly, high level of ROS plays a central role in I/R. During I/R, hydrogen peroxide (H2O2) is the most plentiful form of ROS and cause inflammation, apoptosis and following tissue damages. In this study, we developed H2O2-responsive antioxidant polymeric prodrug of vanillin, termed poly(vanillin oxalate) (PVO) which is composed of antioxidant and anti-inflammatory vanillin and H2O2-responsive peroxalate ester linkages covalently in its backbone. Vanillin, released from PVO nanoparticles, exerts antioxidant and anti-apoptotic activities both in a cell culture model and an animal model of hepatic I/R. Therefore, we anticipate that PVO nanoparticles have tremendous potential as therapeutic and protective agents for I/R injury and H2O2-associated diseases.
