제목 Direct translational analysis using electrophoretically separated DNA in a cell-free protein synthesis system
초록 Cell-free protein synthesis (CFPS) has emerged as a powerful platform for biotechnology and synthetic biology which employs isolated protein synthesis machinery. Cell-free protein synthesis is not subject to the constraint of cell growth and offers greater speed and configurational flexibility in the expression of proteins encoded by target DNAs and assay of protein function.
For the development of a rapid platform for translational analysis of genetic sequences, we attempted in this study to combine the techniques of gel electrophoresis and cell-free protein synthesis in a streamlined manner. After an electrophoretic separation, DNAs in the sliced gel discs were then brought in contact with protein synthesis machinery in the reaction mixture. In the experiments performed with superfolder green fluorescence protein and Candida antarctica lipase B as model proteins, we could successfully translate electrophoretically separated DNAs into functional proteins in a matter of hours.
