Graphene oxide (GO)-based optical biosensor consisting of fluorescent GO covalently linked with a peptide-quencher complex, was developed for the simple, rapid, and sensitive detection of protease activity. The quenching efficiency of various candidate quenchers for GO fluorescence, such as metalloprotoporphyrins and QXL570 , was examined, and their quenching mechanism was investigated. The constructed GO–peptide–QXL system was able to sensitively detect a specific protease like chymotrypsin or matrix metalloproteinase-2 (MMP-2) via a “turn-on” response of quenched GO fluorescence after proteolytic cleavage of QXL from the GO surface. Finally, the GO–peptide–QXL hybrid was able to successfully detect MMP-2 in vitro. |
