Various types of coating polymers have been developed and used for prevention of restenosis with quite success for a long time. In recent years, it receives attraction to develop the system which releases drugs with proper response to stents-surrounding changes to prevent restenosis arising out of inhibiting re-endothelialization due to the unnecessary release of drugs. Among various molecules in vivo, it is known that the Matrix Metalloproteinase-2 (MMP-2) is up-regulated by vascular injury and stent implantation. Therefore, this work has demonstrated the development of MMP-2 crosslinked multi-arm polycaprolactone as a stent coating materials. Multi-arm polycaprolactone was synthesized by ring-opening polymerization with macro-initiator, Boltorn H40, and then it is crosslinked with MMP-2 by p-nitrophenyl chloroformate chemistry. The general characterization was done by 1H NMR and GPC. |
