The cell-instructed delivery system was designed to control the release behavior, according to the cell traction force by tuning the mechanical properties of the cell delivery carrier. First, alginate molecules were substituted with methacrylates and mixed with pure alginate to prepare microgel crosslinked with Ca2+ ions. Then, the interpenetrating covalent cross-links were incorporated in an ionically cross-linked gel. Controlling the amount of chemical cross-links tuned the microgel with varying of stiffness and toughness. Finally, the stem cells were introduced in the microgel functionalized with sulfates molecules for recapitulating the heparin structure. The stem cells differentiated were hatching and released in the functional alginate microgel with the designated work to fracture and elastic modulus. Overall, this results will be useful in designing cell-instructed delivery by systematically controlling the mechanical properties of the gel and the activity of growth factors. |
