The advent of next generation sequencing and mass spectrometry technologies has allowed to determine antibody repertoires at an unprecedented depth. In this talk, I will present two approaches that employed NGS and MS for investigating antibody repertoire. First, I will introduce the high-throughput sequencing coupled with mass spectrometry of serological antibodies. This proteogenomic approach allowed us to quantitatively characterize the influenza-specific serological antibody repertoire. Second, I will discuss antibody sequencing to determine the naïve and antigen experienced repertoires of healthy human donors by using a recently developed large-scale experimental method for pairing heavy and light chain variable regions. These two approaches for antibody repertoire will provide an advance in studying antibody response to disease or to vaccination, aiding the development of therapeutic antibodies. |
