Polycarbonates are well-known biocompatible materials owing to their biodegradable character by hydrolysis, biocompatibility and easy to modify their functional groups. Herein, we described synthetic method of cationic moiety containing polycarbonate with well-defined narrow molecular weight distribution by organocatalytic ring-opening polymerization and the potential as a gene delivery vector was demonstrated. Amine functionalized methoxy poly(ethyleneglycol) was used as a macro initiator of ring-opening polymerization and enhanced particle stability in blood circulation. The cationic pendent groups, such as quaternary and tertiary amines, were introduced for effective complexation with DNA and facilitated endosomal escape. Formation of complex and size distribution of polymer/DNA complex with varied N/P ratio was determined by electrophoresis and dynamic light scattering, respectively. |
