A facile method to prepare the PVA stabilized F-68/heparin/chitosan nanoparticles complex is designed and characterized for protein delivery. F-68 and heparin forms by hydrogen bonding in water, and this mixture formed complex nanoparticles by electrostatic interaction with positively charged chitosan. TGF-ß2 and BMP 7 utilized cartilage regeneration, a model protein, was loaded before the formation of complex nanoparticles by chitosan. FE-SEM was used to observe the formation of PVA stabilized F-68/heparin/chitosan nanoparticles complex and ELS was used to observe the size distribution and zeta-potential. The amount of released TGF-ß2 was determined by ELISA. With the optimized complex nanoparticles, sustained release was accomplished during 7days. TGF-ß2 was loaded into nanoparticles complex, and a sustained release was observed maintaining its bioactivity. These indicate that the nanoparticles complex in this study can be utilized as a sustained delivery system for protein drug. |
