제목 | Measurements and Analysis of Lateral Diffusion of Lipopolysaccharide on Model Bio-Membrane using Fluorescence Tracking (CLSM) Method |
초록 | LPS (lipopolysaccharide) of Gram negative bacteria cell membrane, as a natural oligomer, is known to induce initial immune response and play important roles leading to the endotoxic sepsis shock when it streams in human body. But in view of an initial molecule interaction has been not clear up, there is a limitation to account for a sepsis phenomenon. As a fundamental study of single molecule interaction of LPS, observation mobility changes of LPS diffusion behavior on membrane while reaction with extra proteins, we emboded bio-mimetic LPS membrane on mica substrate by LB method. Using FRAP (Fluorescence Recovery After Photobleaching) technique of CLSM (Confocal Laser Scanning Microscope)1, we treated ROI (Region of Interest) photobleaching selectively. As a result of this experiment, we acquired quantification values via LPS diffusion mechanism (charactierization time(tD),mobile fraction(R), diffusion coefficient(D))2. Supplementally, To examine into change of membrane rigidity, we embeded cholesterols of various ratio into model bio membrane and confirmed. To verify of lipid layer properties, we used IR, XPS, AFM image analysis, X-ray scattering. Fig.1 When a region in the fluorescent area is bleached at time t0 the fluorescence decreases from the initial fluorescence . The fluorescence recovers over time by diffusion until it has recovered .
