제목 Development of artificial naringenin riboswitchesby SELEX and in vivo selection
초록 Monitoring the intracellular concentration of metabolites enables facile control of the pathway gene expression and screening of competitive producer strains. Transcription factor-based sensors are often accompanied by the off-target effect and delayed response which results from trans-regulatory mechanism. On the contrary, riboswitch could bypass the aforementioned issues. Here we demonstrate the process of developing riboswitches which specifically sense the intracellular concentration of naringenin by modulating the concentration of supplemented flavonoids and antibiotics in selection process. Selected riboswitches from few rounds of positive and negative selection work at desired operational ranges as designed selection scheme. Finally, we validated the specificity of the riboswitches for naringenin against other flavonoids with analogous structure. We expect that these new riboswitches could be applied to diverse engineerings to screen optimal strain or monitoring metabolites.
