Polymeric hydrogels have been utilized extensively in tissue regenerative medicine. Recently, it is demonstrated that oxygen plays a pivotal role as a signaling molecule in regulating cellular activities. In particular, growing evidence demonstrated that hyperbaric oxygen facilitates wound healing process and tissue regeneration. Herein, we report hyperbaric oxygen-generating (HOG) hydrogels as dynamic acellular matrices for wound healing and tissue regeneration. We synthesized functionalized polymers that can form hydrogel networks via calcium peroxide (CaO2)-mediated oxygen-generating reaction. We demonstrate that HOG hydrogels rapidly generate molecular oxygen up to hyperbaric levels. Interestingly, we noted that the HOG hydrogels enhance cellular activities in vitro and facilitate wound closing with enhanced tissue infiltration and neovascularization in vivo. Taken together, our HOG hydrogels hold a great potential as a dynamic acellular matrix in tissue regenerative medicine. |
