Image guided therapies can provide suitable way for treatment of hepatocellular carcinoma. The intra-arterial treatment is to increase local and targeted drug delivery while minimizing side effects of anti-tumor agents. However conventional TACE still gives insufficient initial drug concentration. Therefore, we suggested the doxorubicin-albumin-nanoparticles-microbubble (tADMB) to improve therapeutic efficiency by initial cavitation under ultrasound contrast. As a result, doxorubicin encapsulation efficiency of 67.2±8.5%. At a low MI of 0.06, the tADMB were visualized in IO. In rabbit tumor model, tADMB increased the therapeutic efficiency with ultrasound, compared to doxorubicin-IO. On TUNEL assay analysis, near total necrosis of tumor and surrounding of liver tissue was envisaged in tADMB-IO with ultrasound group. To sum up,, our study demonstrates that therapeutic efficiency can be dramatically improved by combining ultrasound contrast with intra-arterial tADMB-IO administration. |
