제목 Chitosan-shelled glucose-responsive polymer vesicles for oral delivery of insulin
초록 Chitosan-shelled nanoparticles have been studied as potential oral delivery vehicles for protein drugs due to their stability under low pH conditions and preferential adhesion to the mucus layer of small intestine. We synthetized amphiphilic block copolymers built on a benzoboroxole-containing glucose-responsive block and poly(ethylene glycol) with α-azido group. The resulting block copolymers were further functionalized with oligo-chitosan moeties via Huisgen cycloaddition. The chitosan-functionalized block copolymers self-assembled into glucose-responsive micelles and vesicles, which showed glucose-triggered disassembly under physiologically relevant pH conditions and glucose levels (10 ~ 20 mM). Chitosan-shelled polymer vesicles could protect the encapsulated protein drugs such as insulin from denaturation and enzymatic degradation in the course of oral uptake. Furthermore, the chitosan-shell might enable the glucose-responsive polymer vesicles to adhere to the mucosal surface.
