Modified PAMAM dendrimers with short oligopeptides for early endosomal escape and enhanced gene delivery |

In recent years, non-viral vectors have become a great topic of research in the field of gene therapy. In this study, we conjugated short oligopeptides to PAMAM G4 in order to achieve higher transfection efficiency. Previous reports have shown enhance gene delivery using PAMAM G4-Histidine-Arginine due to its better cell penetration and internalization mechanism. So we synthesized PAMAM G4-HFR, PAMAM G4-FHR and PAMAM G4-FR to evaluate best gene carrier with low toxicity. Physiochemical studies have been performed measuring zeta potentials and mean diameters. DNA condensation has been confirmed by gel retardation assay. Cytotoxicity and transfection efficiency have performed for HeLa and HepG2. Transfection efficiency was found to be efficient. So our results have shown the promising properties of our carriers to achieve higher transfection with negligible cytotoxicity. |
