Cationic liposomes are useful tools for gene delivery, but most of the them are subjected various obstacles such as degradation by DNase, unexpected binding to blood component, immune reaction and toxic effects both in vitro and in vivo. In this study, we reported stabilized plasmid-lipid particles (SPLPs) with enzymatically cleavable oligopeptides. The SPLP consisted of encapsulated plasmid DNA and PEG-corona liposome. This uniform structure of SPLP can overcome limitations cationic liposome system. Also, SPLP with enzymatically cleavable oligopeptide has an ability to improve transfection efficiency that has been reported previously. The GFLG (Gly-Phe-Leu-Gly) sequence is cleaved by the enzyme cathepsin-B and it may promote endosomal escape after intracellular internalization. The encapsulated DNA was analyzed through lysis by TritonX-100 and PicoGreen assay. Furthermore, transfection efficiency and cytotoxicity were determined with luciferase and MTT assays. |
