Mesoporous nanoparticles have emerged as a promising nanocarrier and provided unique features such as high stability, biocompatibility, and large loading capacity. Herein, we describe a novel approach for protein delivery based on mesoporous silica nanoparticles featuring pH-responsive charge-convertible pore walls. The mesopore surface was functionalized with citraconic amide with terminal carboxyl groups. Surface-functionalized nanoparticles could effectively hold positive proteins such as cytochrome c (Cyt c) within the mesopore at body pH and could facilitate the protein release at below endosomal pH, caused by degradation of citraconic amide accompanying a charge conversion from negative to positive.
