제목 Industrial applications of Allium cepa: Pectin extraction, and subsequent production of succinic acid by CRISPR-cpf1 mediated lactate dehydrogenase 1 mutant of Corynebacterium glutamicum
초록 Succinic acid is one of the top value biochemical in bioconversion fields through microbial fermentation from biomass. Pectin is also a high value functional food ingredient widely used in the food and beverage industry. Onion containing diverse polysaccharides and high soluble sugar is a valuable resource in the industrial fields. The homogalacturonan extraction resulted in a 24.8% increase in enzymatic hydrolysis. The hydrolysate, 54.11 mg mL-1 sugars, was converted to 16.78 mg mL-1 succinic acid by Δldh1 mutant of C. glutamicum through CRISPR-cpf1 gene editing system or 22.0 mg mL-1 bioethanol by S. cerevisiae. The pectin extraction removes the structural retardation factor from the onion cell wall and renders the cellulose more accessible to cellulase, leaded to enhanced saccharification. From the results, we conclude that onion is an attractive resource for economical production of biochemicals and biofuel.
