Gene delivery system (GDS) has been attracted as a promising therapeutic method to treat various diseases such as cancer, immunodeficiency, and genetic disorders, although the clinical application of GDS is still restricted by some limitations including short duration, low transfection efficiency, and low accumulation at specific tissue. The aim of present study is to cite a novel targeting molecule, cholecalciferol, which is specifically bound with vitamin receptor over-expressed in ovarian cancer cell. In this study, Ternary gene complex system was prepared by conjugation of branched polyethyleneimine(PEI) with plasmid DNA, and covering of cholecalciferol-grafted poly-L-glutamic acid. This system showed high transfection efficiency by site-specific uptake and anti-tumor activity in vitro and in vivo. These results revealed that ternary gene complex prepared in this study is a promising carrier for successful gene therapy. |
