Target delivery systems with surface-functionalized polymeric nanoparticles, could be avoid non-specific toxicity and enhance therapeutic efficacy, have been spotlighted. We previously verified a RVG-peptide conjugated PLGA nanoaprticle with neural cell specifity would offer therapeutic approach as siRNA delivery tool for gene therapy in the neuronal tumor. Neural tumor bearing mouse model was established in this finding, and then the nanoparticles were injected intratumorally. Its specific binding affinity of the peptide functionalized the nanoparticles for the tumor tissue was identified. We injected intravenously the nanoparticles with the siRNA into the neural tumor bearing mice, and improved tumor retention and anti-tumor efficacy were significantly observed in the mice treated with the functionalized nanoparticles. This approach to the RVG peptide conjugated nanoparticles containing various drugs could be useful for the design of novel neural-specific delivery systems. |
