A taxis is the directed cell migration in response to the spatial inhomogeneity of mechanical(mechano-/duro-taxis), chemical(chemo-/hapto-taxis), or geometrical(contact guidance) properties of surrounding ECM. While a number of previous researchers studied cell migration with a single taxis, in vivo cells simultaneously experience multiple types of spatial inhomogeneity for various physiological or pathological situations, e.g. development, immune response, and cancer metastasis. Here we generated hydrogel cell culture scaffold with the independently controlled spatial inhomogeneity of substrate rigidity(durotaxis) and density of adhesion ligand(haptotaxis), which allows to compare relative strength between the two taxes and to study their cooperative/competitive effects on cell migration. The scaffold is composed of both photo-crosslinkable polyelectrolytes and photo-cleavable peptide sequences, which with UV irradiation pattern secondary crosslinking and RGD peptide, respectively. |
