Development of scaffold-free tissue constructs with anisotropic cellular assembly using micro-patterned cell-adhesive hydrogels |

Structure of tissue plays critical roles in its function, and therefore, many approaches have been developed to re-create native-tissue like architecture with artificial scaffolds in tissue engineering. However, these systems have often been reported to have problems associated with rejection or toxicity. Alternatively, methods of engineering scaffold-free tissue constructs have been considered to generate highly assembled tissue without use of scaffolds. In this study, we developed a novel system for harvesting confluent cell layers with defined tissue structure using biomimetic temperature responsive hydrogels, incorporated with cell-adhesive peptide (RGD) and molded with controlled micro-patterns. We found that layers of assembled muscle cells with anisotropic patterns can be produced by culture of cells on hydrogels and serial temperature change. Our system may hold potential to engineer tissue that can closely mimic native tissue architecture. |
