Vorinostat-incorporated nanoparticles for local delivery into human cholangiocarcinoma |

This study was to evaluate the anticancer activity of vorinostat and vorinostat-incorporated nanoparticles (VNPs) against HuCC-T1 human cholangiocarcinomas. VNPs were fabricated using poly(DL-lactide-co-glycolide)/poly(ethylene glycol) copolymer. VNPs have anticancer activity similar to that of vorinostat in vitro. Both vorinostat and VNPs have equal potency in terms of inhibition of histone deacetylase expression, mutant-p53 suppression, increased p21 expression and PARP/cleaved caspase-3 expression. However, VNPs showed improved antitumor activity against HuCC-T1 cancer cell-bearing mice compared to vorinostat, whereas empty nanoparticles had no effect on tumor growth. The improved anticancer activity of VNPs can be explained by molecular imaging study using near-infrared dye-incorporated nanoparticles, that were intensively accumulated in the tumor region rather than normal one. Our results demonstrate that vorinostat and VNPs exert anticancer activity against HuCC-T1 cholangiocarcinoma. Thus, we suggest that VNPs are a promising candidate for anticancer chemotherapy in cholangiocarcinoma. |
