Injectable, enzymatically degradable and pH/temperature-sensitive polycarbonate based hydrogel for protein delivery |

Polycarbonates are one of the most widely used materials in biomedical applications due to excellent biocompatibility, high performance, and absence of acidic degradation products that can be harmful to incorporated drugs or proteins as well as in vivo tissues. In this study, we introduced a novel injectable and pH/temperature-sensitive multiblock copolymer hydrogel composed of poly(ethylene glycol) (PEG) and poly(amino carbonate urethane) (PACU) for protein delivery. The copolymer was synthesized and characterized by 1H NMR and gel permeation chromatography. An aqueous copolymer solution quickly formed an in situ hydrogel after subcutaneously injecting into a SD rat. The non-cytotoxicity of the hydrogel was confirmed by a MTT assay and the in vivo degradation investigation was performed on SD rats. The hydrogel has been utilized as depot for sustained release of human growth hormone. |
