제목 | Practical production of acetaminophen via Cu-catalyzed reactions |
초록 | Acetaminophen is a common over-the-counter analgesic for minor pains, such as headaches and is also prescribed as an antipyretic. In spite of its extensive medical applications, any of the developed regioselective process has not been replaced the conventional process that contains nitration of phenol and purification of the desired p-regioisomer. Herein, a potent industrial process for acetaminophen is presented from p-dihalobenzenes via two Cu-catalyzed reactions. Each of the reactions proceeds to introduce the N-acetylamido and the hydroxyl functional groups without the use of the strong acids (HNO3, H2SO4) in the conventional process. The new Cu-catalyzed process regioselectively affords N-acetyl-p-aminophenol in more than 65% overall yields. |
