Small interfering siRNA (siRNA) has recently attracted a lot of attention in the medical field for its considerable potential as an approach to treat various cancers and gene-related diseases, because of the highly efficient and specific silencing of target mRNA expression at the post-transcriptional level. Furthermore, siRNA is a promising approach to the preparation of nanostructures via self-assembly. We have demonstrated a novel strategy to prepare a new class of biologically active siRNA-based nanostructured materials, dendrimer-like siRNA, for superior gene silencing. Dendrimer-like siRNA formed compact and stable polyelectrolyte complexes with biocompatible low-molecular weight linear PEI (LPEI, Mw 2,500) due to its substantially enhanced charge density, compared to naked siRNA. |
