Nanocarriers confer a higher ratio of surface area to volume to improve the pharmacokinetics and biodistribution of therapeutic agents, thus maximize loading efficiency of the therapeutic agents and advanced release profiles. In this study, we developed novel nanocarriers of open porous silica nanoparticles to achieve the purpose of prolonged concentration of therapeutic agents and controlled release. Since our silica nanoparticles have radially oriented mesopores, in which their size gradually increases while proceeding from the center to outer surface of the nanoparticle, they provide large surface area and permit target drugs to easily access the active site of the surface that are located the pores. Thus, the silica nanoparticles are capable of containing a much higher concentration of target drugs than other porous nanoparticles. When dexamethasone and ibuprofen were selected and tested as a therapeutic agent, the loading efficiency into the silica nanoparticles was determined to be ca. 270 %. Furthermore, through surface treatments of the silica nanoparticles, pH and temperature dependent controlled release of the drugs was achieved. |
