제목 Metabolic engineering of Klebsiella pneumoniae for the production of 1,3-propanediol from glucose
초록 1,3-Propanediol (1,3-PDO) is an important platform chemical and its production using recombinant Escherichia coli has been developed and commercialized by DuPont, but the strain cannot produce coenzyme B12. This study aims to develop a more economical microbial cell factory using Klebsiella pneumoniae J2B which can naturally synthesize coenzyme B12. The K. pneumoniae was engineered to have the synthetic pathway for the production of glycerol and modified for improved 1,3-PDO production by numerous iterative design-build-test (D-B-T) cycles. More than 25 genes were modified or overexpressed during the development. The high yield of 0.47 g 1,3-PDO/g glucose was observed.
