제목 Engineering isolated Pseudomonas putida strain for production of 3-hydroxypropionic acid from glycerol
초록 In the present study, to use glucose as a growth carbon source, Pseudomonas putida was developed as host for 3-HP production. The P. putida C3 which can grow well on glucose was isolated, and 3-HP production pathway was heterologously introduced. To enhance the yield and titer of 3-HP, 3-HP degradation pathway and a putative alcohol dehydrogenase converting 3-hydroxypropionaldehyde to 1,3-propanediol were disrupted. In addition, transcriptional regulator GlpR and glycerol kinase GlpK were removed and 3-HP could be produced at the theoretical yield of 1 mol/mol glycerol. On the flask scale, 65 mM 3-HP was produced in 24 h.
