Ratiometric Fluorescent Probe Based on a Peptide Receptor with Femtomolar Affinity for Cu(I) in Aqueous Buffered Solutions |
초록 |
Fluorescent probes based on novel peptide-based receptors for the detection of Cu(I) were synthesized and characterized. Among them, pyrene-labeled tripeptide probe (Py-WPH) formed a 2:1 complex with Cu(I) and had a femtomolar binding affinity (0.3 fM) for Cu(I) from a competition experiment with bathocuproine disulfonate. Py-WPH detected Cu(I) selectively by a ratiometric response among various biologically relevant metal ions, including Cu(II) and Zn(II). The binding mode study showed that the imidazole group of the peptide receptor plays a critical role in potent binding with Cu(I) and the pi-electrons of indole group of the probe might cooperate to form a complex with Cu(I) The fluorescent peptide-based probe penetrated the living RKO cells and successfully detected intracellular Cu(I) by a ratiometric response. This fluorescent probe based on peptide-based receptor with potent binding affinity for Cu(I) will be a potential tool for better understanding of copper biochemistry. |
