With great advances in cancer therapy using polymeric micelles (PMs), the surface-functionalization of PMs with various targeting ligands has been significantly dealt to achieve the enhanced therapeutic efficacy for clinical use. Additionally, the formation of PMs by self-assembly enables to result in its multifunctionality by simply mixing each component, which has the individual targeting moiety. In present study, vinyl sulfone (VS) conjugated PEG-PLLA diblock copolymer as a basic component to form dual ligands-tethered PMs with a nano-size has been synthesized and characterized. Firstly, monotosylated PEG 3.4K (mtPEG) was prepared by using silver (I) oxide and potassium iodide at 0 ℃ for 1 h. Then, VS conjugation to the hydroxyl group of mtPEG was followed by the removal process of tosyl group with Mg-MeOH and ring-opening polymerization of L-lactide. Micellar properties of VS-PEG-PLLA in water were investigated, as well as its physicochemical characteristics. |
