A di-block copolymer, poly(L-lysine)-poly(ethylene glycol) (PLL-PEG) conjugated with folate (FOL) was synthesized. To improve a cancer specific-cellular transfer efficiency of proteins or nonoparticles, the PLL-PEG-FOL conjugate was used as a carrier in a folate receptor-mediated intracellular endocytosis. PLL-PEG-FOL/protein or nanoparticle complexes could be easily obtained due to the ionic interacting between positively charged PLL of the conjugate and protein or nanoparticles having negative surface charges. The PLL-PEG-FOL/protein (nanoparticle) complexes showed enhanced intracellular uptake by cancer cells (folate receptor overexpressed cell line, KB cells). The formulation of the intracellular protein (nanoparticle) delivery system using the synthetic di-block copolymer, PLL-PEG-FOL conjugate, could be also effectively used to folate receptor-mediated endocytosis of other bio-functional macromolecules. |
