제목 In vitro chondrogenesis of rabbit mesenchymal stem cells in hydrogel using tissue engineering technique
초록 In this study, mesenchymal stem cells (MSCs) from rabbits were encapsulated in a fibrin hydrogel and cultured with or without transforming growth factor-b3 (TGF-b3)-loaded nanoparticles to study the potential for chondrogenesis in a hydrogel scaffold system amenable to minimally invasive implantation. MSCs proliferated well in the hydrogels with TGF-b3 loaded in nanoparticles and TGF-b3 alone compared to TGF-b3-unloaded nanoparticles or fibrin hydrogel alone (scaffold disappeared). Immunohistochemistry revealed that large amounts of type II and decreased collagen type I existed in the TGF-b3 loaded in nanoparticles. Upregulation of aggrecan and type II collagen gene expression compared with the absence of TGF-3 was demonstrated. Type I collagen gene expression decreased from 1 to 4 weeks in the presence of TGF-3. This study demonstrates the ability to encapsulate MSCs with sustained release of growth factors in order to form cartilage-like tissue in vitro in fibrin hydrogel.
