Chitosan is one of the useful natural polymeric biomaterials. Poly(N-isopropylacrylamide) is well known to have thermoresponsibility. Poly(NIPAAm) has the lower critical solution temperature(LCST, ca 32℃) at which a macromolecular transition from a hydrophilic to a hydrophobic structure occurs. In this study, thermosensitive chitosan has been prepared by grafting N-isopropylacrylamide(NIPAAm) to water-soluble chitosan(WSC) using ceric ammonium nitrate(CAN) as the initiator. Their reaction chemistry and properties of the graft polymers were investigated. Synthesized graft copolymers were characterized by FT-IR, 1H-NMR, DSC and TGA. The WSC-g-NIPAAm hydrogel exhibited obvious thermal sensitivity. Cell culture was carried out to investigate the biocompatibility of the WSC-g-NIPAAm copolymer. This thermosensitive and water-soluble chitosan might be useful for biomedical and pharmaceutical applications, especially as a injectable material for cell delivery system.
