For economic feasibility of microbial fermentation processes, the utilization of abundant and cheap carbon source is essential. In this regard, acetate is one of the promising carbon source which can be obtained from various sources as by-product or waste. In this study, we engineered the Escherichia coli strain to produce L-tyrosine from acetate. First, the linear pathway from acetate to L-tyrosine was amplified with synthetic biology based tools. Thereafter, the glyoxylate shunt was optimized for enhanced L-tyrosine production by varied expression of isocitrate lyase. The significantly increased L-tyrosine production was validated in resulting strain. These efforts support that acetate can be a potential feedstock for biochemical production with engineered E. coli. |
