Preparation and Characterization of Chitosan-DNA Complex for introduction into a Gene Delivery System |

The ability of the gene condensation was investigated by gel retardation assay. LMWSC with free-amine group was formed the complex at the low weight ratio, above a 1:4 weight ratio of plasmid DNA : LMWSC. However, COS-DNA complex was accomplished above 1:8 because of lactic acid bound at the amine group of chitosan. Particle sizes of LMWSC-DNA and COS-DNA were about 131.0 nm and 193.1 nm, respectively. Because LMWSC with free amine group has the strong positive charge, it can be formed the compact complex with plasmid DNA. The stability of complexes was retained above 10 days. In cytotoxicity test, all WSC and its derivatives complexes showed negligible cytotoxicity for 293T cells and HeLa cell. |
