All-Trans Retinoic Acid Release from Polyion Complex Micelles of Methoxy Poly (ethylene glycol)-grafted-Chitosan |

We synthesized methoxy poly(ethylene glycol)-grafted chitosan (ChitoPEG) copolymer to prepare RA-encapsulated polymeric micelle. RA-encapsulated polymeric micelle of ChitoPEG copolymer is 100-500nm in particle size and has spherical shape when observed by TEM. In 1H NMR study, the specific peaks of RA and chitosan as a drug carrying innercore disappeared at D2O and only the specific peak of MPEG was observed, while specific peaks of MPEG, RA, andchitosan appeared at DMSO. This indicates that RA/ChitoPEG ion complexes are composed of polymeric micelle with a core-shell structure, and that free drug does not exist on the polymeric micelle formulations. Another evidence of drug-incorporation into polymeric micelle was witnessed in a DSC analysis. The melting peaks of RA and chitosan were 182 oC and 220 oC, respectively. |
